Additional Information: | The study aimed to detect early maturity and genetic diversity of mutant lines of sugar palm (Arenga pinnata Merr.) accessions originating from Banten induced by gamma ray. A total of 10 SSR primers specialized to detect early maturity of sugar palm was used to assess the molecular level of total mutant lines along with early maturing (Kutim) and late maturing (Toumuung) palm sugar varieties as controls. This research was conducted by quantitative and qualitative methods. The main steps of this study are genomic DNA isolation, quantitative and qualitative DNA tests, dilution of the DNA, DNA amplification by PCR technique, electrophoresis of PCR results with 6% polyacrylamide, scoring of DNA bands and data analysis. The results showed the genetic variation of the gamma rays induced mutants of local sugar palm accessions from Banten compared to the accession plant without radiation. The individuals plants of the early maturing sugar palm of Kutim tend to close with mutant lines from Serang with dose 10 gy, Lebak with dose 7,5 gy, and Pandeglang with dose 5 gy, indicating their early maturity. This SSR markers demonstrated that the mutants of Banten's local palm plants revealed low genetic diversity. The genetic distance based on similarity of the mutant group suggest their high similarity to the control sugar palm plant (without radiation) with a value of 0.97. The genetic distance among mutant lines and control ranged from 0.99 to 0.75. This genetic diversity and identified early maturing mutant lines are important information which could be beneficial for the genetic improvement of sugar palm in Indonesia.
