Additional Information: Drought can cause declining of rice production, to develop drought tolerant rice lines with ideal agronomy and high yields that has been conducted through the backcross breeding program between Situ Bagendit as the recipient and Cabacu as a donor parent. The current backcross population has reached the BC3F2 generation carrying the drought-tolerance QTLs (qRPF2.1 and qGPP2.1) from Cabacu. These individuals need to be selected by phenotype and genotype analysis (foreground) using microsatellite markers. The objectives of this study were to determine the phenotype performance of the BC3F2 rice line for drought tolerance, know the agronomic characteristics of the BC3F2 rice line when compared to its parents, genotype condition of the BC3F2 Situ Bagendit × Cabacu rice line based on microsatellite markers, and obtain BC3F2 Situ Bagendit × Cabacu rice lines which have phenotypes and genotypes suitable for drought tolerance. This research was conducted at the Laboratory of Molecular Biology and the Cimanggu Greenhouse of the Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development (BB-Biogen) from October 2019 to May 2020. The result of the phenotype and genotype selection indicated that Situ Bagendit × Cabacu BC3F2 for drought tolerance obtained two individuals who had phenotype results with the drought-tolerant category, the agronomic characteristics following of Situ Bagendit and Cabacu, had the desired QTLs (qGPP2.1 and qRPF2. 1) was F15.3 - 17 and F15.3 - 21. The selected plant can be used to develop the next breeding generation of Situ Bagendit × Cabacu rice lines.
