Abstract

The aim of this research is to determine the effect of folic acid and the various light intensities on the growth of patchoulis explants (Pogostemon cablin Benth.) by using in vitro method. This research was conducted in April to July 2016 in the Biotechnology Laboratory, Faculty of Agriculture, Sultan Ageng Tirtayasa University Serang, Banten. This research used Block Randomized Design, which consist of two factors. The first factor was a concentration of folic acid consisting of 4 levels, there were : A0 (0 mg/l), A1 (0,25 mg/l), A2 (0,5 mg/l), A3 (0,75 mg/l). The second factors was the light intensity which consist of 3 levels, there were : B0 (76 lux), B1 (1156 lux), B2 (1818 lux). There were 12 combinations of treatments, each treatment was repeated 4 times, so that there were 48 units of the experiment. The result showed the treatment of folic acid concentration did not give any effects on the parameters of time comes shoot, number of root, plant height addition and number of leave and the parameters of number of shoot gave significant effect. Treatment of various light intensities gave significant effect on the number of shoot, plant height addition parameter at 1, 2 week after plant and the number of leave on 8 week after plant. There were interaction between the concentration of folic acid and various light intensities on addition parameters plant height at 1, 2, and 3 week after plant, and the number of shoot. In conclusion, the use of a combination of folic acid and various light intensities A3B1 (folic acid 0,75 mg/l + light intensity 1156 lux) was the best combination for the number of shoot.
