Abstract

GITA NUR RIZAL. In Vitro Tissue Culture Response of Modification MS Media and The Concentration of PEG 6000 towards Growth of Patchouli Explants (Pogostemon cablin Benth.) With guidance by Susiyanti and Moh. Ana Syabana. This research purpose to determine the effect of modification of MS media and PEG concentration on the growth of patchouli explants (Pogostemon cablin Benth.) with in vitro method. This research was conducted in March to June 2015 in the Biotechnology Laboratory, Faculty of Agriculture, Sultan Agung Tirtayasa University Serang, Banten. This research used a completely randomized design (CRD), which consists of two factors. The first factor is a modified MS media consisting of 4 levels, there are: M0 (¼ MS), M1 (½ MS), M2 (¾ MS) and M3 (1 MS). The second factor is the concentration of PEG which consists of 4 levels, there are: P0 (0%), P1 (2%), P2 (4%) and P3 (6%). There are 16 combinations of treatments, each treatment was repeated 2 times in duplicate, so that there are 64 units of the experiment. The results showed the treatment of MS media modified was not given effect on the parameters of plant height addition, number of leaves has given significant effect on 6 and 7 MST, and the parameters of the number of shoots was not given significant effect. Treatment of different concentrations of PEG 6000 has given significant effect on plant height addition parameter at 5,6,7, and 8 MST, the number of leaves on 3,4,5,6,7 and 8 MST and the number of shoots on 8 MST. There were interaction between the modified MS media and the concentration of PEG 6000 on addition parameters plant height at 6 and 8 MST, number of leaves at 4,5,6,7, and 8 MST and the number of shoots on 8 MST. From the results of this research concluded that the use of a combination of all parameters is the best media on patchouli explant in vitro is M2P3 (¾ MS + 6% PEG 6000).
