Abstract

Seaweed is one of the leading commodity fisheries in Indonesia. Seaweed farming success is influenced by the use of seaweed seedlings were superior. The availability of quality seeds can be helped by breeding through tissue culture techniques. The aim of this study was to determine the exact concentration PES media for callus growth Kappaphycus alvarezii seaweed tissue culture. The benefits of this research is to get the composition and concentration of the media PES affecting the growth media. This research was conducted in October to November 2015 at Biotechnology Laboratory of Department of Agroecotechnology Faculty of Agriculture, University of Sultan Ageng Tirtayasa. The research was used the media's treatment of PES different, namely A1 (5 ml/L PES), A2 (10 ml/L PES), A3 (15 ml/L PES), A4 (20 ml/L PES) and A5 (25 ml/L PES) performed four replications each treatment. The parameters in this study is the explants formed callus, percentage of live explants, callus colour, time of callus formation and callus texture form. The results of the study concluded that concentrations of PES appropriate media for callus growth of seaweed Kappaphycus alvarezii A3 treatment media is adding a stock solution of PES 15 ml/L with the result of 75% callus formation time, explants formed callus explant percentage of 75% and 100% live.
