Abstract

Effect of Concentration 2,4-D and Lid Colors on Callus Growth Patchouli (Pogostemon cablin Benth.) In vitro Pogostemon cablin Benth. is a plant that has a high economic value. So it is necessary to develop a method to accelerate the propagation of plantlets. This research aims to determine in vitro effect of the concentration of 2,4 - D and wavelength on the callus growth of patchouli plant (Pogostemon cablin Benth.). The research was conducted from March to May 2016 in the Laboratory of Biotechnology Faculty of Agriculture, University of Sultan Ageng Serang Tirtayasa Serang. The culture medium Murashige and Skoog modified by the addition of 2,4-D hormones. This study used a randomized design (RBD), which consists of two factors. The first factor was 2,4-D concentration consists of four levels : D0 (0 mg /l); D1 (0.5 mg /l); D2 (1 mg/l); D3 (1.5 mg/l); D4 (2 mg/l). The second factor consisted of five levels of illumination color: B0 (control), B1 (red), B2 (yellow), B3 (green), and B4 (blue). The combination of these two factors resulted in 25 combined treatment with four replications of each treatment. Hence obtained 100 experimental unit. The results showed that the addition of 1 mg/l gave the best results in the diameter of callus patchouli. While the convex green plastic gives the best effect on the growth of callus patchouli. There were combination effect on 2,4-D and lid colors concentration on initiation time callus and diameter callus. There was interesting the best effect on callus diameter that is at treatment D4B3 in diameter callus. All the living explants during the observation reached 98% with the number of contaminating explants was 6 % and browning explants 10 %.
