Abstract
This research was aimed to know the effect of growth regulators 2-ip (2-isopentyl adenine) and folic acid to shoots of agarwood plant (Aquilaria malaccensis Lamk.). This research was conducted in August to November 2014 at Biotechnology Laboratory of Sultan Ageng Tirtayasa University, Serang Banten. This research used completely randomized design (CRD) developed in factorials and consists of two factors. First factor is concentration of 2-ip (2-isopentyl adenine), consist of three levels namely: B0 (0 mg /l), B1 (0.5 m /l), B2 (1 m /l). Second factor is concentration of folic acid consist of 5 levels namely: A0 (0 mg/l), A1 (0.25 mg/l), A2 (0.5 mg/l), A3 (0.75 mg/l), A4 (1 mg/ l) and there were 15 treatment combination, every treatment repeated three times and there were 45 experiment units. Observation was conducted every week to parameters namely plants height, shoots number, leaves number, and in the end of observation to parameters namely percentage of explants forming shoots, percentage of explants life, and the percentage of contamination. The results of research showed that the different concentration of 2-ip (2-isopentyl adenine) and folic acid did not give any effect to all parameters of agarwood plant. There is interaction between the concentration of 2-ip (2-isopentyl adenine) and folic acid to the parameters shoots number at age 4 weeks after planting. From the results of research can be inferred that treatment 2-isopentyl adenine (0 mg/l) is the best treatment for the parameters shoots number, a combination of treatment between 2-isopentyl adenine (0 and 0.5mg/l) + folic acid (0.5 mg/l).

Additional Information: | Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh 2-iP (2- Isopentyl Adenin) dan asam folat terhadap tunas tanaman gaharu (Aquilaria malaccensis Lamk.) secara in vitro. Penelitian ini dilakasanakan pada bulan Agustus sampai bulan November 2014 di Laboratoriun Bioteknologi Fakultas Pertanian Universitas Sultan Ageng Tirtayasa Serang Banten. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang disusun secara faktorial dan terdiri atas dua faktor. Faktor pertama konsentrasi 2-iP (2- Isopentyl Adenin), terdiri dari 3 taraf yaitu : B0 (0 mg/l), B1 (0.5 mg/l), B2 (1 mg/l). factor kedua konsentrasi asam folat terdiri dari 5 taraf yaitu : A0 (0 mg/l), A1 (0.25 mg/l), A2 (0.5 mg/l), A3 (0.75 mg/l), A4 (1 mg/l) sehingga terdapat 15 kombinasi perlakuan, setiap perlakuan diulang sebanyak 3 kali sehingga terdapat 45 satuan percobaan. Pengamatan dilakukan setiap minggu untuk parameter tinggi tanaman, jumlah tunas, jumlah daun serta pada akhir pengamatan untuk parameter persentasi eksplan membentuk tunas, persentasi eksplan hidup, dan persentasi kontaminasi. Hasil penelitian menunjukan bahwa perlakuan konsentrasi penggunaan 2-iP (2- Isopentyl adenin) dan asam folat tidak memberikan pengaruh yang nyata terhadap semua parameter tanaman gaharu. Terdapat interaksi antara konsentrasi 2-iP (2- Isopentyl adenine) dan asam folat terhadap parameter jumlah tunas umur 4 MST. Dari hasil penelitian dapat disimpulkan bahwa perlakuan 2-Isopentyl adenine (0 mg/l) perlakuan terbaik untuk parameter jumlah tunas, perlakuan kombinasi antara 2-Isopentyl adenine (0 dan 0.5 mg/l) + asam folat (0.5 mg/l). | |||||||||
