Abstract

HERY MARGATAMA. The Effect of Explant Size and Concentration of PGR 2,4-D with In Vitro Callus Induction of Kappaphycus alvarezii. Supervised by DODI HERMAWAN and SUSIYANTI. Seaweed is a potential biological resource in Indonesia, especially seaweed of Kappaphycus alvarezii. The seeds of seaweed until today still depend on seeds from the nature it self. If this condition still existed, it will cause many problems such as lack of seeds and reducing of seeds qualities. The purpose of this research was to determine the optimum size of the explants and the effect of PGR concentration of 2.4-D on the growth of callus tissue culture of Kappaphycus alvarezii. This research consisted of two factors: the size of the explants and concentration of 2.4-D PGR. The parameters measured of explants colour, colour of callus, callus formation time, percentage of live explants, percentage of explants forming callus, callus texture, and form callus. The results showed the percentage of callus generated 58%, 73% the percentage of live explants, callus texture generated an average crumb to form filaments filamentous callus. Callus formation fastest time average of 14.33 HST on the treatment of explants size 10 mm with the addition of 2.4-D PGR 1 mg/L. Then, the color parameter callus from PGR concentrations of 2.4-D 1 mg/L showed significantly different results is 3.25. Explants color shown in this case resulted in the best value treatment of PGR concentrations of 2.4-D 1 mg/L with 4.62 point. The best results in this research was found in the addition of 2.4-D PGR 1 mg/L. Keywords: callus, explants, Kappaphycus alvarezii, PGR 2.4-D
