Abstract

The aim of this research was to identify Korea rice cultivars conferring resistance to abiotic stresses using specific DNA markers. The research was conducted from May to August 2015, in Laboratory of Molecular Biology, BB-BIOGEN Bogor. A total of 155 rice cultivars consisted of 147 introduced from Korea and eight elite varieties as control plants were amplified using molecular markers related to abiotic stresses, such as drought, saline, aluminium toxicity, and flood tolerance. The result showed among 147 Korean Rice cultivars amplified with three DNA markers related to drought, such as RM20A, RM228 and RM336, we detected a total of 66, 134 and 117 rice genotypes carrying resistance gen to drought, respectively. Regarding the marker linked to saline stress, six out of 147 introduced rice cultivar were identified to have the salinity resistance gen amplified with RM493 marker. Meanwhile, 37 out of 147 rice samples carried allele conferring alumuniun-resistance gene. In the meantime, amplification product of Korea Rice cultivar using AEX markers resulted in a total of 27 rice cultivar carried allele conferring resistance gene to puddle or flooding stress. These findings would be very useful as early step to screen useful genes sourced from introduced rice cultivar for future purpose in breeding program. ABSTRACT De Ratna Melati.
